核酸试纸条法检测结核分枝杆菌的初步研究

Abstract This paper aims to establish a visual nucleic acid strip method for rapid detection of Mycobacterium tuberculosis.Primers and probes for Mycobacterium tuberculosis were designed according to IS6110 gene and were labeled with biotin and FAM.The recombinase-mediatedthermostatic amplification was carried out first, and then the amplification products were detected by dipstick after being diluted. The results were determined according to the quality control line and the test line.The sensitivity, repeatability and specificity of the method were analyzed.The reaction process of amplificationcould be completed at 37℃ , which takes 15 min and strip chromatography was used for the testing of products, which takes 15 min. Results were interpreted by naked eye. The sensitivity of the strip method in our study is 104 copies /μL. The establishment of nucleic acid test strip method is quick and easy to operate, and it is suitable for POCT detection without professional equipment.
